Easy on-chip melting curve measurement
The graphs present normalized melting profile and temperature derivative for 11 pairs of perfect matched (PM) - mismatched probes (MM). The PM probes are in blue and MM probes in red. Melting curves are measured by applying a temperature ramp to the hybridized microarray and recording the fluorescence signal decrease as a function of temperature. For each probe, the background-substracted signal is normalized against its value before heating and further normalized against a reference probe. The resulting melting curve is fitted with two parameters and the first derivative is taken to calculate the respective melting temperatures.
Acquisition and analysis software
HybLive™ comes with a dedicated control and analysis matching software package. It has been designed with versatility and ease-of-use in mind, and the result is a powerful tool that integrates perfectly with the hardware.
Programming an experiment is a matter of seconds thanks to the hybridization and washing protocol templates included? Any parameter and setting can be overridden, should you need finer control over the hybridization process than these templates provide.
Detailed and robust analysis of the sequence of images is easily performed. The results can be plotted on a variety of graphs and exported in various standard formats.
